The mcpB gene product is expressed under conditions that are unknown. The Hughes lab isolated mutants that express McpB when grown on L plates (mcpB). The mcpB locus was later found to be linked to the yodD and salM genes. A three-factor cross was set up to determine the location of mcpB relative to yodD and salM. P22 transducing phage grown on a mcpBoFF salM yodD strain was used to transduce a mepB salM yodD strain to salM. Results are below: Colonies arising on plate Spread on plate Phage only (108 Recipient cells only (10) Mixture of phage and cell:s 468 (10 of each) (a) Why did one colony grow up on the plate with phage only? (b) Why did one colony grow up on the plate with cells only? (c) What is the genotype of colony on phage only plate? d) What is the genotype of colony on cell only plate? Problem continues on back

e) Recombination is a cell process which results in a restructuring the part of a genome. Replica plating is a microbiological technique in which one or more secondary Petri plates containing different solid agar-based selective growth media such as antibiotics are inoculated with the same colonies of microorganisms from a primary plate which replaces the original pattern of colonies.
