In a TLC technique we take a base line on which spots of reactants and reaction mixture are put. The TLC plate is then kept in a TLC chamber in which little amount of solvent is kept to elute the TLC. The base line of the TLC should be always above the solvent. Here, the student has dipped the TLC plate in the solvent above than base line which has resulted in the dissolution of base spots in the solvent, thus, base spots are coming down instead of going up
Below is a picture of a TLC plate by a student using this technique and developing...
1) A student develops their TLC plate and places it under a UV light, but nothing appears. What mistake might the student have made? a) Spotted the compound below the level of eluent b) Spotted the compound above the level of eluent c) Used a sample that was too concentrated d) Used too much sample 2)Thin layer chromatography, TLC, is commonly used to monitor reactions to completion. Using this technique, what would indicate to a student that the reaction is...
5. Two unknown samples are spotted next to each other on a TLC plate. Development of the plate with ethyl acetate and visualization using UV light gives the following data: Spot 1: Rp = 0.65 Spot 2: Rp = 0.63 These two Rf values are so close that the difference might be due to experimental error (you're still learning TLC and not yet an expert in the technique). Describe an experiment that can be run using the same developing solvent...
2. If two different compounds have the same Rr value, how might they be identified using paper chromatography? 3. Why is it important to keep the spots on the paper as small as possible? 4. If the solvent pool at the bottom of the beaker touched the spots on the 1.5cm line, what would happen to the spots? How would you address this error? 5. If you were teaching a student how to do paper chromatography, what three pieces of...
