Question

Cac), Towever, h most applications it is advantageous to store DNA in TE Buffer (10mM Tris with 1mM EDTA). Why is it problematic in this experiment to resuspend the DNA in TE buffer? 6. Assume that a 50 μg/mL solution of native DNA has an A260 of 1, calculate the concentration of your DNA sample in mg/mL. Then, assuming that 10% of the onion cells mass are the nuclei and that 10% of the nuclei mass is DNA, calculate the percent yield of your isolated DNA. (Note: it will be very smal...) 7. Referring to Appendix 2 prepare a sample calculations section to include in your notebook. iscussion tips n addition to including all the required components of a good discussion as outlined in Appendix 2 please consider the following points when writing your discussion: Discuss the hyperchromic effect as seen in your experiment. . Answer #1

(6) A260 of 1. Concentration of DNA sample = 50 μg/mL

Concentration of DNA in mg/mL = 50/1000 = 0.05 mg/mL (Answer)

Concentration of DNA in onion cell = 10% X 10% X 0.05 mg/mL = 0.1 X 0.1 X 0.05 mg/mL = 0.0005 mg/mL
