You’re studying proteins and the effect of pH on those proteins. For that purpose you are going to make two buffers. First, how would you prepare a 2L of a 10 mM Phosphate buffer at pH of 7.4?

You have –

Solid KH2PO

Solid K2HPO4

10M KOH (as much as you need)

purified water

pH meter

balance, glassware and anything else you need.

The three pKa values for phosphoric acid (from the CRC Handbook of Chemistry and Physics) are 2.16, 7.21, and 12.32.

(Note: there is more than one way to do this and you don’t necessarily have to use all of the above.)

Then pick a second buffering agent to use at a different pH. It must be at least 2 pH units away from the buffer above. Explain how you would prepare 2L of 10mM of that buffer as well.
