Tubular reabsorption is the process by which most of the substances from the filtrate are resabsorbed into blood stream .

Water ,amino acid, glucose are resabsorbed in the early proximal convoluted tubule ,but for the reabsorption to take place it needs sodium gradient maintenance means sodium is prevented to build up in the epithelial cells of the tubule and it is achieved by sodium /potassium ATPase.

Chloride ions are resabsorbed in the late proximal convoluted tubule. They are resabsorbed by chloride/anion exchange.

So, if a drug blocks the reabsorption of sodium then it will also block the reabsorption of water, chloride, amino acids and glucose . Hence there will be disturbance in the osmotic pressure and will effect the extracellular body fluid volume.
