Grams stain is used to test the bacteria either it grams positive or negative and to observe it's shape.

Most bacteria stain either blue (grampositive) or red (gram negative).

The cell walls of gram-negative bacteria have a thin layer of peptidoglycan and a high lipid concentration that allows alcohol to penetrate and leach out the crystal violet-iodine complex.

Those organisms then take up the red counterstain.

Gram positive bacteria has thick cell wall which doesn't allow alcohol, so that it remain s blue.

Bacteria are classified into 5 groups based on basic shapes:

  • spherical (cocci)
  • Rod (bacilli)
  • spiral (spirilla)
  • comma (vibrios) or
  • Corkscrew (spirochaetes).

If we observe the above slide

Bacteria absorbed blue and it is in rod shape.

So that slide shows gram positive bacilli.
