One glucose molecule catabolised into two pyruvate molecules through 10 step reaction process known as Glycolysis.

Glucose is a Six Carbon molecule and Pyruvate is a 3 carbon molecule.

In the 4th step of Glycolysis ( when Fructose-1,6-bisphosphate to Dihydroxy acetone phosphate and Glyceralde-3-phosphate ) the six carbon atom chain break into two 3-carbon atom molecule.

One pyruvate molecule will have carbon atoms C1,C2 and C3 and other will have Carbon atoms C4,C5 and C6.

So, 50% of pyruvate molecules will have labeled carbon isotope 14C.
