4. A researcher has developed three different cell lines that are able to each ignore one of the checkpoints (G1, G2 or M) in the cell cycle. Develop a test (by adding or taking away certain chemicals) that could be used to distinguish and separate the three different cell lines?

Ans) For studying the cell cycle, the cells have to be synchronized i.e they all should be in the same phase of the cell cycle.

Suppose all the cell lines are initially in the Go (resting phase).

1) We will remove serum from the cell culture in cell line 1. We will observe that the cell line 1 will be arrested at Go phase.

In cell line 2 and 3, we have not removed serum from the medium and thus cell lines 2 and 3 will proceed to the next phase of cell cycle i.e G1 phase.

2) Similarly, to arrest at S phase, we add Hydroxyurea (which inhibit DNA replication) to one of the cell line culture.

3) To arrest at M phase, we add nocodazole (which disrupt spindle).

By this way, we can arrest different cell lines at different phases of cell cycle to study them one by one.
