A very popular drug in the treatment of the herpes simplex virus (HSV-1) is called acyclovir. When it comes to the treatment of HIV, the virus that causes AIDS, the most popular drug of choice is AZT (Retrovir).
A) The nucleoside analogies contains the nucleic acid analogue , a phosphate moeity and a Sugar molecule . Addition of the nucleotide analogues result in the termination of the process of replication . It also competes with the nucleoside to participate in the process of replication. Therefore it act as a competitive inhibitor of nucleoside. It also results in the process of termination. Acyclovir is a purine analogue that is adenosine and Guanosine analogues structure . AZT or Zidovudine is used in termination of HIV replication . AZT works on the mechanism of the salvage pathway in which it is converted into monophosphate form .
B) Summary :- DNA
replication is inhibited due to the the absence of 3'OH at the 3' end .The monophosphate form is converted into triphosphate form which binds to the the DNA in the replication process and inhibits the the further replication . The mechanisms is well explained in diagram .

The methyl group in thymidine has been replaced by an iodine atom 12.3.1 Nucleoside analogues Antiviral therapy is aimed at preventing viral DNA from being replicated within the host cell. The strategy is to use molecules which resemble natural nucleosides but which are modified to make them nonfunctional. These are called mucleoside analogues. (Case 7) "THE HIV" Aja is a 24-year-old seeking prenatal care as she is one month pregnant with her first child. Her prenatal HIV antibody screen results positive for Human Immunodeficiency Virus-1. Aja reveals that she has a history of multiple sexual partners and no former antiviral therapy. Aja's bloodwork shows that her CD4 helper T-cell lymphocyte levels are normal, however, treatment with the antiviral drug AZT (also known as Zidovudine) is prescribed to aid in prevention of passing the...
