Washed and unwashed skin differs in their total microbial load. Washing the skin with water removes the surface bacteria into the water.

The use of a scrub brush removes more bacteria from the skin than with water alone. This might be due to the removal of bacteria from the lower layers of the skin as well in response to mechanical scrubbing action.
