What would you expect to happen to a newly synthesized soluble
protein if a mutation caused the deletion of the KDEL
sequence?
a. The protein would be returned to the ER in COPI-coated
vesicles.
b. The protein will be carried forward through the Golgi
apparatus.
c. The protein will be trapped in COPII vesicles .
d. The protein will be incorporated into COPI vesicles.
e. The protein will be recognized by KDEL receptors and a retrieval
sequence (KDEL sequence) will be added to it.
After synthesis most of the soluble or non-soluble proteins reach to Golgi. Golgi apparatus decides the next destination.
Proteins synthesized for the lumen of endoplasmic reticulum are recognized by the KDEL sequence present at their carboxyl terminal. This is called ER retrival signal. Mutation in this signal does not allow the protein to bind with KDEL receptor present in Golgi. As a result the protein can't return to endoplasmic reticulum and are carried forward through the Golgi apparatus.

please answer all that you can 1. You have genetically engineered green fluorescent protein (GFP) containing a KDEL sequence (GFP-KDEL). When GFP-KDEL is expressed in normal human fibroblasts and examined using fluorescence microscopy, the fluorescence appears diffuse across the cytoplasm. How would you explain this observations given that KDEL is supposed to be an ER-specific sorting sequence? A. This engineered GFP would not have a hydrophobic signal sequence to get it into the RER in the first place. B. The...
4) A cell line is transfected with a plasmid encoding either a secreted glycoprotein, X, or a variant of X that contains the amino acids KDEL at its C terminus, both of them under control of an inducible promoter, KDEL motifs are normally found on proteins located in the lumen of the ER. At 30 minutes after induction, cycloheximide, an inhibitor of protein synthesis, is added to the culture medium. At 30, 45, and 60 minutes after induction (that is,...

5. Consider the wild-type F8 gene. What would happen to the 1) transcript (mRNA) sequence and quantity, 2) to the protein (sequence, quantity, function) and 3) the person’s overall phenotype if they were homozygous (both copies of the gene are the same) for a 3 base-pair deletion in the: i. First intron ii. First exon iii. The promoter iv. 5’UTR v. Last exon 6. What environmental factor could affect the phenotype caused by an F8 null mutation? Thrombophilia is a...
