Barfoed reagent is a mixture of copper acetate and acetic acid. The test is based on reduction of Cu2+ to Cu2O (red precipitate) by monosaccharides. Disaccharides are much weaker reducing agents hence they do not reduce Cu2+ under the weakly acidic test conditions. The redaldehyde group in the monosaccharide gets oxidized to acid.
