Stem-loop or also known as hairpin loop is intramolecular base paring that occurs in single stranded DNA. This stem loop is formed due to complementary nucleotide base pairing within same DNA strand .This stem loop is usually found at the 5'UTR(Untranslated region) of prokaryotes. These structures cause the attenuation of a transcript in order to regulate translation.This loop are generally 8 to 10 base pair long. So if we remove the termination stem-loop sequence in a prokaryotic genes then the mRNA formed will not be released and protein formation will not take place (translation).Stem loop are rich in cytosine and guanine base pairs which has three hydrogen bond and are quite strong to break.The stem-loop structure is important in prokaryotes for Rho-independent transcription termination.
