In order to determine if mutation from different organism that exhibit the same phenotype are allelic, Complementation test would have been performed.

A complementation test can be used to test the mutations in two strains are in different genes, complementation will not occcur if the mutations are in same gene.

The convinience and essence of this test is that the mutations that produce a phenotype can be assigned to different genes without the exact knowledge.

Example- suppose two strains of white -eyed flies of the species Drosophila melanogaster,commonly known as fruitfly.In this species , wild type flies have red eyes and eye colour is known to be related to 2 genes,A & B. Each one of these genes has two alleles, a dominant one that codesfor a working protein (A&B) and a recessive one (a&b) that codes for a malfunctioning protein. Both the proteins are necessary for red pigmentation in the eyes. If a given fly is homozygous for either a&b , it will have white eyes.
