In this experiment, we sought to measure glycogen and free glucose in tissue homogenates, using spectrophotometry to detect levels of glucose in tissue samples. We inferred the amount of glucose stored in glycogen using reaction catalyzed by amyloglucosidase. A control reaction was without amyloglucosidase, this measured free glucose or not part of glycogen.
If we fail to vortex before taking out glucose or glycogen then concentration of glucose and glycogen will be less that what actually it should be as there is gradient of concentration of glucose or glycogen formed from top to bottom.
More concentrated glucose or glycogen are present at base while at top its conc. Is less.
Glucose and glycogen both attached to water and become heavy and settled down at base.
