In methyl- directed mismatch repair the strand that is repaired is not the methylated strand. It is unmethylated strand that is repaired.

MutS is the mismatch recognition protein. It has ATPase activity to beak the bond and remove the mismatch. It recognizes base-base mismatch, deletions, insertions etc in the DNA molecule. MutL binds to MutS and together activate MutH.

DNA pol is needed to replace the correct nucleotides.
