As T cell progenitors (thymocytes) progress through lineage development in the thymus, their expression of CD 44 and CD25 changes. Draw a dot-plot of CD44 vs CD25 expression divided into quadrants, separating high and low expression of these proteins. Indicate on the dot-plot where you’d expect DN2, DN3 and DN4 thymocytes to be located (Use a labeled circle for each). Explain your reasoning.

Answer #1

DN1 cells are CD44+CD25- which give rise to DN2 population. DN2 progresses to DN3 and then finally to DN4 pupulation.

DN2 is CD44+CD25+(UN 2) 一个h54) Co 25 -

DN3 is CD25+CD44–

DN4 is CD44–CD25–
