Answer #1

The basic difference between ion exchange and gel filteration chromatography is that ion exchange works on the mechanism of their affinity to bind to ion exchanger and gel filteration chromatography works on the principle of seperating the amino acids or prteins on basis of their size and shape as they eluted from the matrix. In case of Ala, Val, Gly, Tyr, Arg, Val, Lys, Ser the best method to distinguish between them is by gel filteration method as they have considerable differences in their molecular weight we are able to elute them on the basis of their size and mass. The seperation is done as small proteins are embeded into gel beads and larger proteins are eluted first and only a small amount of mobile phase is used in the elution process. After that small proteins will be eluted. The order of elution is following:

Tyr>Arg>Lyc>Val>Ser>Ala>Gly
