Answer #1

Describe the mechanism responsible for pharmacodynamic opiate tolerance (explain how beta-arrestin is involved)

Answer:-Opiate Tolerance is process that causes reduced opioid effect.Use of opioid for long period causes ceiling of receptor which leads to opiate tolerance. It is of 3 types Pharmacodynamic,Pharmacokinetic and behavioral.Causes of pharmacodynamic tolerance

1)High concentration of a substance constantly binding with the receptor desensitizing it through constant inetraction.

2)Reduction in receptor density or other mechanism

3)Increased receptor density or other mechanism

Beta Arrestin 2 is G-protein-coupled receptor causes desensitization through interaction of the opiate receptor which develops opiate tolerance. It is type of pharmacodynamic so it is known as Pharmacodynamic opiate tolerance,
