You inoculate M. caffeinum into both caffeine and MM-sucrose at 8 AM. Because this species is filamentous, its growth is most accurately measured dry weight. At regular intervals throughout the day, by 10 mL filter the cells onto you vigorously mix the culture, remove weigh the disk. M.caffenium would grow better on MM- sucrose , because inspite of reaching exponential phase at the same time in both the media, the bacteria grown in MM-sucrose has more dry weight at exponential phase as compare to dry weight of bacterium grown in MM-caffeine.
