"In one model/diagram, show how transciption factors, histone modifiers, chromatin remodelers, DNA methylation all cooperate to regulate the thermostat for an individual gene (how much mRNA is transcribed)."

Lets say you've discovered a gene tht codes for a protein that interferes with your ability to scratch-off quiz cards. Clearly you need to shut this gene off.

You can name your gene and its regulatory transciption factor(s) whatever you want, but the rest of your model should be accurate. Make sure to include at least one specific example from each of the categories of regulation listed above (hint-they are not necessarialy listed in the order you will need to use them).

The promoter for this gene contains several CpG islands-this is where you diagram should start...

Could anyone draw a diagram AND please explain it. My teacher had her baby early and our class is completely lost and the sub has no idea (not his fault, he is a plant dude).

In vertebrates, approx 70-75% gene promoters are associated with a CpG, making this the most common promoter type. it controls most of the housekeeping genes and some tissue-specific genes. they also make transcription start sequence.

mammalian transcription binding sequence are richer in CpG and many contain CpG in their recognition sequence. for example like Sp1 transcription factor.

also in few cases where gene are active due to some kind of inducer, there are CpG sequence where nucleosome is less (Ramirez-Carrozzi et al. 2009). this is due to methylation of histone proteins that binds to DNA. H3K4me3 is a signature histone mark of CGI promoters.
