Question

Can someone explain why methionine is not added to the plate?

Consider the cross: Hfr: strS arg+ met+ gal+lac+lys- x F-: strS arg- met- gal-lac- lys+ where the genetic map is shown below (the < indicates the Origin of transfer in the Hfr strain).

a.

Minimal media + galactose + streptomycin + arginine + lysine + methionine + lactose

b.

Minimal media + galactose + arginine + methionine + lactose

c.

Minimal media + galactose + arginine + lysine

d.

Minimal media + galactose+ arginine + lactose

e.

Minimal media + streptomycine+ arginine + lysine + lactose

f.

Minimal media + galactose+ arginine + methionine

g.

More than one of these above

h.

None of these above

Which media would allow the growth of the Hfr strain?

The correct answer is c.

I see the correct answer, but I do not know why methionine is not included in the plate. I thought we leave amino acids on the plate if we are not trying to kill the parental
