The most commonly used DNA sequencing method was Sanger sequencing also known as the chain termination method sequencing. Sanger sequencing comes in two forms, manual and automated. One of the big difference between these method is the means by which reaction products are detected. In manual sequencing reaction products are detected by using dideoxy nucleotides and in the automated sequencing reaction products are detected by fluorescent labelling or autoradiography.
