1. You are tasked with examining a new exciting protein. This protein is membrane bound but can be cleaved and detected in the serum. Where within the spleen is the protein located? Describe how you’ll experimentally determine this in details. Comparatively speaking is there more of this protein before or after infection with Francisella tularensis in the spleen?

Answer:          This question i have to write pointswise, becuase it's easy to understand you...

  • The given protein is expected to be present in the outer lining of the tissues which are in contact with the tissues
  • As it is a membrane-bound protein with its cleavage site outside the cell. Hence, it is easy for the cleavage of the protein and its presence in the serum.
  • Before the infection with Francisella tularensis, there will be a normal concentration of protein, however, at the time of infection, the protein concentration will reach its peak due to damage to the spleen tissue.
  • Hence, after infection, there would be a minimum concentration of protein in the serum.
  • Hence for the detection of protein concentration, We will be using the ELISA technique with the specific antibodies A and B.
  • Antibodies A and B are chosen because of their closely related detection sites.
