DNA Sequencing Techniques

Compare/contrast Sanger sequencing, pact-bio, and oxford nanopore sequencing techniques. Include how to make a library, and why you would use each one in respect to cost, size, % error, etc. Consider budget, what you want to get out of it, and is there merit to combine different technologies together?

Answer #1

Sanger sequencing was the first sequencing method developed by Frederick Sanger. It is a method of first generation sequencing whereas pact-bio and Oxford nanopore are third generation sequencing methods.

Comparison of these methods are

Sanger sequencing Bio-pact Oxford nanopore
Cost of sequencing Highly cost High cost than Sanger and Oxford

Low cost

Size of genome get sequence in a one run 300-1000 bp 500000 bp 10000-10000bp
Time for sequencing Require high time Requires least time than other two methods Required medium time than other two methids
Accuracy Very high 99 percent

98 percent

From all these methods, one can get the genome sequence OR part of genome to perform various research activities. Yes, there is a merit in combining these methods so that we can perform sequencing techniques at very low cost along with minimum time
