Why would you silence gene expression for both wild-type and mutants? I am on a question that asks why a researcher would use shRNA to reduce the expression of endogenous SRP on both wild-type and mutant SRPs. What would it help test for translational pausing in co-translational import?

During an experiment silencing of wild type gene is done to know that if this gene is not found in the genome of an organism, what are the effects of this knock out gene is found in organism.  

Mutant gene is also silenced to know that if there is another gene in the plant which produces result in organism or it such mutant gene are expressing in that manner in organism.

snRNA is also known as small nuclear RNA. This snRNA has some sequences complementary to the SRP of wild type and mutant gene in cytoplasm, this snRNA bonds with gene and leads to double stranded mRNA. Argonauts protein acts on this double stranded RNA and cleaves into small pieces. Hence their expression is reduced.

If we take snRNA has sequence complementary to wild type and mutant , you will find that there will be no cotranslational import. But if you take snRNA sequence to either one of the wild type or mutant type, then there is only one protein either mutant or wild type undergo translational import. And co-op translational import of wild and mutant type is paused.

If there is no snRNA, then you will find do translational import in both cases.
