Reduced physical activity is the main cause of increase in weight in these models resulting in energy imbalance. Orexin signaling promotes obesity resistance via enhanced spontaneous physical activity and energy expenditure regulation. Lack of orexin causes increase in the weight.

Potential adverse effect will be obesity, depression, learning and memory deficit.
