Procedure for performing a urinary eosinophil count in AIN :-

- Collect a random urine specimen of about 10 ml.

- Spin 10ml of a random urine collection in centrifuge machine.

-Pour off supernant .Assess urine sediment for the presence of white blood cells(WBC).If no WBC are seen ,the eosinophil test can't be performed.If WBC are present ,prepare slide using spun sediment.

--Hansel Strain Method is preferred to count the number of eosinophils in urinary sediment after straining with Hansel strain which is a mixture of methylene blue and eosin Y.

-Microscopic examination of strained slide for counting the number of eosinophils.

- A Positive result has been defined as greater than 1% eosinophils but a result of greater than 5% is more specific for Acute Interstitial Nephritis(AIN).
