Retention factor (Rf) can be known from the TLC using a suitable solvent mixture (example: hexane + ethyl acetate) as an eluent, by which it can be confirmed that which compound in the mixture will be eluted first from the column.

Hence, it is necessary to perform TLC before carrying out the column chromatography.
