Cetuximab is an epidermal growth factor receptor (EGFR) inhibitor used for the treatment of metastatic colorectal cancer, metastatic non-small cell lung cancer and head and neck cancer. Cetuximab is a chimeric (mouse/human) monoclonal antibody given by intravenous infusion that is distributed under the trade name Erbitux in the U.S. and Canada by the drug company Eli Lilly and Co. The initial dose is 400 mg/m2 administered over 120 minutes and Cetuximab is available in 100 mL vials containing 200 mg of drug.

(a) Calculate the initial dose for a patient weighing 140-lb and 65 inches in height.

(b) Calculate the volume of product needed to meet the dose.

(c) The package insert warns that the infusion rate should not exceed 10 mg/minute. If the infusion is given over 120 minutes, is this compliant with the warning?

A. The intial dose for the patient is 660.4 mg.

B. 330.2 ml of volume of product is needed to meet the dose.

C. For this particular patient the required initial dose is 660.4 mg. If the drug is given over 10mg \ min then 66.04 min is required to complete the dose. It may used as warning in over height patients but it is not a warning in this particular patient.
