1. Transformation is the process of inculcating the vector containing the gene of interest into a bacterial cell by spreading 1microlitre of the inoculum on the LB agar plate in the Laminar Air Flow. The vector inserted in the plasmid cells have a ampicillin resistant gene and galactosidase gene, if the DNA of interest in inserted than it dis orients the open reading frame of the vector and thus the dye X-GAL is not able to express on the medium containing the colonies. Also the E.coli cells carry a massive quality of resistance towards ampicillin.
