The suspension loaded on to hemocytometer is assumed to be uniform suspension. However, in practice, the cells may not be uniformly present as single cells. Cells can be clumped. The counting of four separate square will prevent the bias or error that may result. The average number of cells obtained from the 4 squares will give a better representation of what the actual cell count is.

The large squares also allow counting of cells easier. The border gridlines also help to decide whether to count the cell or not.

It is asked not to count the actual % viability in the table 15.1. This is because the % viability is calculated from the average number of cells and not from individual cell count.

Average number of cells= Adding Total number of cells from each square/4

Total number of cells in each square= Dead cell+ viable cell

Total number of cells in square 1= 1+6=7

Total number of cells in square 2= 3+4= 7

Total number of cells in square 3= 1+ 3= 4

Total number of cells in square 4= 2+4= 6

Average number of cells= (7+7+4+6)/4= 24/4= 6

Please note that the 1.75 that is shown is average number of viable cells in suspension (1+3+1+2/4=7/4= 1.75). This is not average number of cells in suspension.

Answer should be 6.
