2. Rolling and Theta circle mode of bacterial replication : Rolling circle is an unidirectional mode of replication, which initiates by breaking of single strand DNA/RNA by DNA polymerase III at 5 prime end whereas Theta replication occurs by breaking at replication fork end of DNA at two positions. Rolling circle mode will creates multiple genome from a single break of DNA whereas Theta mode will gives a new genome inside the older one. In rolling circle, leading strand and lagging strand will covalently attach together to form concatamers, in theta model, topoisomerase II enzyme will seperate the new strand from older one to form seperate catenanes by decatanation process.
