There are plenty types of bacteria present in the soil which provide a perfect site to judge the antibiotic resistance in the organism at normal environmental conditions in a PARE project to demonstrate the mechanism in the students. Therefore, its shows the suitability to investigate the antibiotic resistance in bacteria. We need to dilute the antibiotics and check sub lethal effects of the antibiotics in organism and later develop resistance.

For identification of the organism, one need to have appropriate number of organism, so, it is pertinent to choose the well-formed and established colony to determination.

The 16 s rRNA is a conserved housekeeping gene present in the microbes and conserved and utilize as tool for the identification of bacteria. This gene is endogenously expresses and when compare 16 S ribosomal RNA sequence of two organisms then no changes in the expression levels can be expected (at normal condition, and generally un-altered other conditions too and more than 96% homology between two organism.
