When immunodetection is combined with electrophoretic seperation of proteins the technique is called immunoelectrophoresis. It involves the separation and identification of proteins or antigens based on differences in electrical charge and reactivity with antibodies or immunoglobulins.

The support medium used in this technique is the agarose gel, as it is has no charge aof itself and unreactive. The antigen mixture is placed in wells made in agarose gel plate .When electric current is applied to the gel plate, the antigen mixture starts migrating towards respective electrodes depending to their size and charge. Those having negative charge migrate towards positive electrode, and those with positive charge moves towards negative electrode. Once the antigens get separated, the gel plate is removed from electrophoretic tank and the antibody solution is added to the trough made in the gel and then left for sometime so that diffusion can take place. During this period, antigen and antibody diffuse in all directions and towards each other in the gel plate., They interact and form precipitation lines at the respective zone of equivalance. Curved precipitation lines are formed indicating antigen antibody interaction. Mainly commassie brilliant blue is used as protein stain but the precipitation lines so formed may be enhanced by staining with Amido black stain.

The advantage of immunoelectrophoresis is that different antigens can be identified based on the intensity , shape and position of the precipitation curves and helps in the qualitative analysis of serum immunoglobulins.
