In the experiment regarding transformation, the plasmid containing ampicillin resistance is transformed into normal E.coli cell. This enables the bacteria to survive in the presence of ampicillin. Hence this is practiced to screen transformed bacterias from the non-transformed ones. If they are killed by ampicillin then there has been no transformation. Arabinose sugar production is used for induction of a specific gene not in bacterial transformation.

Bacterial plasmid is not the only genetic material in bacteria. The bacterial chromosome is also present in bacteria as a compulsory genetic material. Moreover plasmids are absent in many bacterias. They only provide additional genetic roles and are not definitely housekeeping.
