This plot is called Lineweaver Burk and it is the perfect graph to visualize inhibition type in an enzymatic reaction. The point at which the line crosses the Y axis is equal to 1/Vmax, and the point where is crosses the X axis is equal to -1/Km. This means we can completely visualize any change in Vmax and Km with this graph.
When we add indol to the reaction we can see how Vmax is not altered but Km actually IS. This pattern is related to COMPETITIVE inhibition, and this means indol interacts with the enzyme by binding to the same active site used for the substrate, that is why it is "competing" with the substrate. The same Vmax can be reached but you need a lot more of substrate concentration to reach it, that is why Km is increased.
a) Competitive inhibition
b) Vmax stays the same while Km increases
c) Indole binds to the active site in the enzyme (not allosteric sites)
a. Identify the type of inhibition occurring. Explain how you know. b. Describe the difference or...
10.What type of inhibitor is this? How do you know? (2)
11.For your assigned inhibitor 1, what are the apparent Km &
Vmax? (NOTE: apparent Km& Vmax are just the Km & Vmax in
presence of inhibitor, at a given concentration.) (2)
Kinetics experiments were performed on PGI. Enzyme activity
(initial velocity, Vo) was measured at varying concentrations of
Glucose-6-phosphate (G6P). The enzyme concentration used in all
experiments was 1.5 μM.

reference. also, hypothesize what will occur in the presence of an
inhibitor and the type of inhibition.

Case Study Diabetes Mellitus Type I You may work with a partner or group and hand in one case study for the group. Make sure all names are on the case study. Do not “divide and conquer” – work together. All answers require a reference you ½ point will be deducted per question. The Lewis text book is a good resource. But also utilize your Maternal Child as well as your nutrition text. These may be helpful. Scenario You work in...

Stroke
A stroke is serious, just like a heart attack, so it's important
to know the signs of stroke and act quickly if you suspect someone
is having one. Stroke is the fourth leading cause of death in the
United States, and causes more serious long-term disabilities than
any other disease. Older people are at higher risk. You can take
steps to lower your chance of having a stroke.
