Question

t cells that are wildtype for the genes in the adenine pathway are of growing on minimal media lacking adenine. In parallel experiments, we O treat a wildtype culture of yeast cells with one of three mutagens: nitrous acid (a deaminating agent), UV light, and the intercalating agent, acridine orange. We obtain several adenine deficient colonies [ade- with each of the treatments and one mutant from each treatment condition is chosen for further analysis. DNA sequence analysis reveals that each mutant contains a mutation in the same adenine gene. However, a computer virus corrupts the data and we are unable to match molecular biology, we turn to you to deduce which treatment corresponds with the sequence with the mutagen treatment. With your expertise in the mutant sequence. Strain DNA sequence type of mutation Mutagen (transition, transversion deletion, insertion) Wildtype ATTCGGCTATA wildtype No treatment Mutant 1 AD6CGGCTATA Mutant 2 ATICGGTATA Mutant 3 ATTCGGCTACAArsiun a. Answer #1

1) in mutant 1 - one transition and one transversion are there. this is due to the intercalating agent acridine orange.

2) in mutant 2 - there is a deletion of C which might be due to deamination with nitrous oxide.

3) in mutant 3 - there is transition of T to C. this is due to UV radiation.
