As part of the regulatory response to decreasing oxygen levels, the FNR protein of E. coli upregulates expression of the Fumarate and Nitrate Reductases, as well as the pyruvate formate-lyase enzyme. What is the function of the pyruvate formate-lyase enzyme, and why would it be up-regulated under anaerobic conditions?

Pyruvate formse lyase is an important enzyme in anaerobic glucose metabolism in bacteria. Using radical non redox chemistry it is responsible for converting pyruvate and coenzyme A into acetyl co enzyme A. it is an iron dependent protein which is required to activate PFL – pyruvate formate lyase.

It is upregulated in anaerobic conditions because it is a highly intracellular oxygen level dependent enzyme. It contains oxygen sensitive radical that is very critical for its activity. If it is exposed to oxygen it will affect the catalytic activity and abolish it.
