ELISA( Enzyme-linked immunosorbent assay ) test is a serology test in which COVID 19 antibodies are tested in the serum sample. ALISA test is microplates based enzyme immunoassay technique used for rapid testing of victims.

In this test, the sample is added to microtiter in microplate coated with COVID 19 peptide antigen nucleocapsid protein. Unbound protein matrix removed from microplate by subsequent washing after the primary incubation period. Bounded protein are treated for subsequent operations. Horseradish peroxidase tracer antibody is added in each well. After the second incubation period immune complex if COVID 19 polypeptide antigen forms if their coronavirus antibody is present in the testing sample.

The unbound antibodies are again washed from well. HRP label tracer antibody is again incubated for a predefined time with substrate solution and measure in a spectrophotometer microplate reader. The enzymatic activity of the tracer antibody is proportional to the COVID 19 antibody present in the sample that is detected directly.
