1.- What is the purpose of using antibodies in the ELISA test?

The antibody use allows the test to be extremely specific for the antigen in question, that is the target antigen.

2.- In the last step of performing the test should the enzyme or the antigen or the substrate be added to the walls??

In the last step the substrate is the added molecule. We already have the antigen and the enzyme in the wall, we only need to add the substrate to trigger the reaction of possibly attached enzymes. This reaction is the one that gives the signal we read to evaluate the presence of the antigen in the sample
