The correct option  that lists the 4 steps of ELISA in order :-

(C) Antibody absorbed to well> Sample antigen added and binds to antibody> Enzyme-linked antibody added to well> Enzyme substrate added and causes color change.

ELISA is Enzyme Linked Immunosorbent Assay. ELISA detect either antigen or antibodies or haptens in the specimen by using enzyme-substrate system.

ELISA is usually performed on a microcicer place containing 96 wells.

Step 1 :- Antibody is absorbed in the well.

Step 2 :- Sample antigen is added which binds to the antibody in the well.

Step 3 :- Enzyme Linked antibody is added to the well.

Step 4 :- Enzyme substrate added and causes color change.

The enzyme reacts with the substrate, which in turn activates the chromogen to produce a color.

The color change is detected by spectrophotometry in an ELISA reader. Intensity of the color is directly proportional to the amount of detection molecule (Ag or Ab) present in test serum.

The other options are incorrect since the order is wrong. In options (b) & (d), the last step which is the change is colour, is placed earlier. Also in option (a) the steps order are incorrect.

Thus the correct answer is (c) Antibody absorbed to well> Sample antigen added and binds to antibody> Enzyme-linked antibody added to well> Enzyme substrate added and causes color change.
