RNAase is the enzyme commonly used in the DNA extraction protocols during the isolation of plasmid. This enzyme is used to remove the bacterial RNA during the plasmid isolation. RNA can contaminate the plasmid that can be seen during the gel electrophoresis. RNAase catalyze the degradation of the RNA into smaller components.

RNAase bind specifically to the RNA. This enzyme binds to RNA in a sequence specific manner. DNase is also a type of nuclease enzyme. DNase catalyze the hydrolytic cleavage of the phosphodiester linkage present in the DNA backbone as a result degrade the DNA. So, if the DNase instead of RNase will be present in the solution it will harm the plasmid and whole experiment will be a waste.
