Fresh,high quality water is used in the preparation of the media. The high quality water is prepared by the process of,

  • Distillation
  • De-ionisation
  • Reverse osmosis

The above methods are directed for the satisfactory results for the constitutions of the media culture. Tap water is not used for fbe preparation of media because the tap water contains impurities like calcium and magnesium and also their metal ion traces. Sometimes,the characteristics of selective media is altered by the chlorine and fluoride.

The places which having hard water should be softened before the process of distillation. The boilers also leads to the contamination of media during autoclaving.so,condenser boiler distillate should be avoided. The tap water shows variable nature in its content , the water should maintained in a specific range of PH. And the use of metal containers with pipe work will leads to the metallic ions into the water and which will leads to the contamination.so these all are to be avoived during the preparation of media.
