In many cellular membrane systems, the V-ATPases (vacuolar ATPases) functions as proton pumps. The V1 portion of the V-ATPase which is hydrophilic in nature is a rotary motor that has a central axis DF complex that rotates inside a catalytic A3B3 complex which is hexagonally arranged by using ATP hydrolysis energy.

The tight binding between the A3/B3 complex and DF complex results in the stable ATPase activity of the complex and the synthesis of the ATP. The loose binding between them results in reduced activity of the ATPase.
