In this scenario, in the absence of the when the cofactor is bound to the repressor, the operon is switched off. Only when cofactor is released, the operon is expressed. In a repressor mutant, unable to bind the cofactor, no repressor is bound to the operon and expression is constitutive.

This requirement of cofactor to bind to the repressor molecule is a defining feature of Negative Repressible operons, where the operon is repressed in the presence of a cofactor, while whn the cofator is absent, repressor is not boun to the DNA and expression is 'on'.
