  1. The enzyme urease is present.
  2. Urea is hydrolysed.
  3. The agar medium is now alkaline.

Urease hydrolyses the urea into CO2 and ammonia. Ammonia alkalinises the medium thereby changing the color from yellow (pH 6.8) to bright pink (pH 8.1). This color/pH change can be usually measured by phenol red.
