PAP or poly-A polymerase is a template-independent polymerase that belongs to the DNA polymerase beta (polĪ²) family of enzymes that catalyzes the addition of about 200 -250 adenine residues at the end of m RNA called poly-A tail that protects the m RNA from degradation.

The capping process replaces the triphosphate group with another structure called the "cap". The cap is added by the enzyme guanyl transferase.  In the reaction, the beta phosphate of the RNA transcript displaces a pyrophosphate group at the 5' position of the GTP molecule.

Incoming aminoacyl-tRNAs (a tRNA with an amino acid covalently attached is called an aminoacyl-tRNA) enter the ribosome at the A site. Initiator t RNA which carries methionine in eukaryotes enters P site directly. Rest of the amino acids bounced to t RNA enters through A site.

The aa-tRNA delivery step is catalyzed by the eukaryotic Elongation Factor 1 (eEF1) complex.
