An Ames test is used to find mutagenic effect of a chemical.

It uses salmonella typhimurium or Ecoli.

Point mutation in histidine or tryptophan operon.

So that they are incapable of producing corresponding aminoacid.

If chemical is mutagenic it will reverse aminoacid prototrophy.

This is reverse mutation.

These reverted bacteria start producing histidine and they can grow well producing more colonies

The colonies in control is due to spontaneous mutation.

Here the colonies of control and test are similar 2 and 3.
