Here, control E.coli cells are used to study transformation. These cells lack the genes which will provide them with ampicillin resistance and also genes encoding beta galactosidase that can break down X-Gal. Therefore they cannot grow on LBA+amp (plates containing ampicillin) and LBA+amp+X-Gal (plates containing ampicillin and X-Gal).
Only after successful transformation, the genes responsible for ampicillin resistance and beta galactosidase will be provided to the E.coli cells and they will be able to grow on LBA+amp and LBA+amp+X-gal.
